What is Birinapant?

Birinapant (TL32711, TL-32711) is a peptide mimetic of mitochondrial pro apoptotic protein SMAC and a potent antagonist of XIAP and cIAP1, with Kd values of 45 nM and less than 1 nM, respectively. It can induce the self ubiquitination and proteasomal degradation of cIAP1 and cIAP2 in intact cells, thereby forming RIPK1: caspase-8 complex, activating caspase-8 and inducing tumor cell death. TL32711 targets cIAP associated with TRAF2 and eliminates TNF induced NF- κ B activation.

Biological activity

Birinaant (TL32711) is a SChemicalbookMAC mimetic antagonist that is most effective against cIAP1. In cell-free assays, Kd is less than 1nM and its effect on XIAP is weak. it can help induce cell apoptosis in HIV-1 infection. Phase 2.

In vitro research

The Kd of Birinapant combined with XIAP and CIAP1 is 45nM and<1nM, respectively. In TRAIL insensitive SUM190 (overexpression of ErbB2) cells, it alone induces cell death (IC50 of approximately 300nM) and significantly increases TRAIL induced apoptosis in TRAIL sensitive SUM149 (triple negative, EGFR activated) cells. It leads to rapid degradation of CIAP1, activation of caspase, cleavage of PARP, and NF- κ Activation of B. TL-32711 and TNF- α Combination therapy exhibits strong in vitro anti melanoma effects. TL 32711 and TNF- α The combination of (1ng/mL) inhibits the growth of human melanoma cell lines WTH202, WM793B, WM1366, and WM164 with IC50 values of 1.8, 2.5, 7.9, and 9nM, respectively, while both compounds alone are ineffective. it alone can induce inhibitory effects on WM9 cell proliferation, with an IC50 of 2.4nM. it significantly inhibits target proteins CIAP1 and cIAP2 in these cell lines.
