What is Acitretin API?

Acitretin API, is a drug made from vitamin A, also known as all-trans retinoic acid or betarretin A. It controls how many skin cells proliferate and differentiate while also affecting how the stratum corneum develops. It improves disorders like keratosis, lessens acne, and controls skin pigmentation by attaching to the vitamin A acid receptor in the nucleus of cells and changing gene transcription and protein expression. With a molecular weight of 326.43 g/mol and a chemical formula of C21H26O3, It often takes the form of a yellowish crystal or crystalline powder. It has the CAS number 55079-83-9. Hexane can be used to solidify it, and its melting point is 228 – 230 °C. The LD50 by intraperitoneal injection ranges from> 4000 mg/ kg( 1 day) to 700 mg/ kg( 10 days and 20 days), according to exploration on acute toxin in mice. Function & Application

Acitretin API is an oral medication belonging to the retinoid class of drugs. It is primarily used to treat various skin conditions, particularly severe psoriasis (severe plaque psoriasis), keratinization disorders, and palmoplantar keratoderma.

• Psoriasis treatment: it reduces the itching, scaling, red spots, and joint pain associated with psoriasis. It controls cellular division and proliferation, which lessens excessive keratinization and inflammatory reaction and lessens the severity of psoriasis.

• Treatment of keratinization disorders: it can be used to treat loricrin keratoderma and non-epidermolytic palmoplantar keratoderma. The symptoms and look of the skin are improved by reducing excessive skin keratinization.

• Treatment of palmoplantar keratoderma: it can treat palmoplantar keratoderma by reducing the thickness and level of keratinization in the palms and soles, alleviating symptoms such pain, fissures, and increasing keratinization.

Production method

4-methoxy-2,3,6-trimethylbenzaldehyde (Ⅰ) and ethyl formate react in phosphoric acid-containing ethanol to yield compound (Ⅱ) with a yield of 84%. Then, it reacts with (Ⅲ) in ethyl acetate containing zinc chloride to yield a mixture of 6:4 (E)-(Ⅳ) and (Z)-(Ⅳ), with a total yield of 83%. Acid elimination gives 27% of (E,E)-(V) and 21% of (Z,E)-(V). (E,E)-(V) reacts with acetone in a methanol-water solution to yield 25% of compound (Ⅵ). It reacts with ethoxyethynylmagnesium bromide Grignard reagent in ether to yield compound (Ⅶ) with a yield of 48%. Finally, hydrolysis yields a 91% product.
